About Camptothecin

Camptothecin and its active derivatives inhibit DNA topoisomerase I (Topo I). Topo I normally creates single‑strand breaks to relieve torsional stress during replication and transcription; CPT stabilizes the Topo I–DNA cleavable complex, preventing re‑ligation. Collisions of replication forks with these stabilized complexes produce DNA double‑strand breaks and cytotoxicity, particularly in proliferating cells.
